Tumor Suppression after Tumor Cell-targeted IL-8 Gene Transfer

Abstract
To study the effect of localized secretion of chemokines on tumor growth, the gene of human (hu)-IL-8(interleukin 8), was introduced, via mammalian expression vector, into Chinese hamster overy (CHO) cells, and the ability of transfected cells to form tumors in vivo was evaluated. The production of hu-IL-8 by transfected clone consistently did not change the growth rate in vitro, but drastically suppressed tumor growth when injected subcutaneously(s.c.) to nude mice. Histological xamination
at the site of injection of CHO clone transfected with hu-IL-8 showed a predominantly neutrophilic infiltration. These results indicate that IL-8 has a potent antitumor activities when released, even at low dose, at the tumor site, which seems to be mediated by recruiment and targeting of neutrophilic granulocytes to IL-8-releasing cells. Our studies highlight the potential usefulness of localized hemokinesecretion to induce potent antitumor immune responses.
